Can someone tell me if I have already sent in Statement in support of claim and now have a VA file # do I put this VA file #
in section 1 of form 21-534? I am ready to send in my Application but I am not sure if this correct.

If the VA has assigned a file number, then yes, use it on the form.
Make sure to keep a complete copy of the application for your records, send it Return Receipt and make sure you mail to the proper Pension Center for your mom's state.
Good luck with the process.
